The star of early 2000s show The OC will make a guest appearance alongside Neighbours stalwarts and some new faces in the new era of the show.
Ms Barton will play Reece, an American new to Erinsborough who is not quite who she appears to be.
"I'm excited to be part of this iconic show's next chapter, and I am really looking forward to being back in Australia - a place I know and love," she said.
"I think the character of Reece is going to be a great role for me to explore and play with."
The long-running soap was given a lifeline by Amazon Freevee after the final episode aired in late 2022.
The show announced the reboot had started production on April 17.
Neighbours' executive producer Jason Herbison said the show was "beyond excited" to have Ms Barton join its new chapter.
"Mischa's character is dynamic and unpredictable, and will have an instant presence on Ramsay Street," he said.
"Our loyal viewers are going to love her, locally and abroad."
Producers announced Annie Jones (as Jane Harris), Rebekah Elmaloglou (Terese Willis), Georgie Stone (Mackenzie Hargreaves), and Tim Kano (Leo Tanaka) will return in their roles as series regulars.
They join the previously confirmed returning soap icons Stefan Dennis (Paul Robinson), Alan Fletcher (Karl Kennedy), Ryan Moloney (Toadie Rebecchi), and Jackie Woodburne (Susan Kennedy).
