Emergency services were called to Fossickers Way, about 8km out of Manilla towards Tamworth after the crumpled wreckage of a Holden Astra was discovered.
The impact of the crash felled the tree, which blocked part of the road, Ambulance NSW said.
Paramedics and police scoured the area about 2.45am, but found no trace of the driver or any occupants.
Officers then returned during daylight on Friday morning and searched the nearby area.
